Just got a Maestro and going to put it in my modified FLSun 300x300x400 printer.
With my 12v power supply, do I need to configure or move the jumpers for the fans? The stock jumper set up is for 24v, but if only 12v power, do I need to do anything?

The fan jumper will either supply 5v, or VIN power. So if you have a 12v supply, it will provide 12v, as there is no way to get 24v from 12v.
