Newb question here. I would like to use the Duet WiFi on the second iteration Lulzbot AO101. I figure if I upgrade the card and the hot end and get it working I will then be able to print most materials. It does have a heated bed an even though it's a small footprint I thought it would be a great test until I build a larger one. What you all think, is this doable?

I'd think that was perfectly do-able. It's effectively MendelMax/Prusa i2 derived printer; see https://reprap.org/wiki/LulzBot/AO-101. Classic! I always like the MendelMax type printers, and you should have a nice little printer once updated.

Use @Phaedrux's very useful guide to the Ender 3 as a starting point/guide to understanding what's involved. See: https://duet3d.dozuki.com/Guide/Ender+3+Pro+and+Duet+Maestro+Guide+Part+1:+Wiring/37
I doubt there's anything that would be odd/difficult about updating this printer. Update it with a probe, for ease of use and levelling capabilities. https://duet3d.dozuki.com/Wiki/Choosing_a_Z_probe

And you could always get longer extrusion, rods, and a larger bed if you wanted to scale it up in the future.
