I am not aware of anything in this area that needs fixing. Please post your config.g file.

            HI
Is this a bug or do I need to reconfigure something to get 1.21 to work with 2 independent (z and E1) motors/drivers

I can't see any problem with your config.g file. My guess is that only one of the Z motors has switched from x16 to x64 microstepping. Does it help if you re-send the M350 command again later in config.g?

It works fine in 1.20…should i define the other motor as U axis etc...?

In your M584 command you must define the new extruder drive assignments too. See the description of M584 on the GCodes wiki page.
