Are you powering the Duet from USB power only? If so then there may be insufficient power from the USB port on your PC or laptop to power the Duet + PanelDue.

Given the communication between the Duet and the PanelDue is serial it wouldn't affect the screen like you describe.

          How is it connecteded?

You can try disconnecting the PanelDue from the Duet and applying USB power directly to the PanelDue. If you still get the vertical lines, try erasing and re-flashing the firmware.

Does that mean you have tried powering the Duet by USB? I am suggesting that you try powering the PanelDue from its own USB connector, to see if there is an issue with power coming from the Duet.
