After fully configuring my CR-10 S properly, my Duet board dropped wifi signal and refused to send a reading through usb connection.

Each stepper motor and end stop where functional before, the hot end and bed where working (but not tested to 220c / 50c yet) the BL touch was also working and everything was updated to Duet 3.2. Basically everything was checked individually and the configuration was complete. The board was correctly enclosed with no loose wire or potential metal dust.

Then, after configuring the z-offset of the BLtouch, we try to heat the hotend to 220c and the board "crashed", no wifi, no USB connection anymore.

Now, the 3.3v led is not on anymore (with the input voltage or usb). The board was never connected with input and usb connection at the same time. Now, when the board turned on, the BL touch does its startup routine but no wifi, no usb and no 3.3v led / esp8266 led. The Input voltage was tested and shows a 12.06v which seems to be pretty correct.

doesn't sound very promising, but try disconnecting everything that uses the 3.3v (sd card, end stops, etc) and see https://duet3d.dozuki.com/Wiki/What_to_do_if_your_Duet_won't_respond

Is this a brand new board? When and where was it purchased?

When going through the link bearer provided check if any of the chips gets hot when using only USB power.
