    This might be a dumb question.. I've upgraded my Ender 3 with a Duet 2 Wifi board and seeing how I'm going from Marlin to RepRap firmware, I was wondering if I need to tell my slicer, Simplify 3d in my case, that my printer is now using a different firmware..

      Ideally, yes. You should set the gcode flavour to RepRapFirmware. This usually tells the slicer to use the gcode syntax particular to your firmware.

      I don't use S3D, but maybe this will help.

      If you're using recent firmware for the Duet, it will accept the common Marlin gcodes as well as the RepRap versions.
