I have a Duet 2 Maestro board running on an Ender 3, I am thinking of adding another Z axis stepper motor and screw rod, I see that the control board has an additional plugin point so that I can run the existing and the new stepper motor in series.

In my config.g file I am supplying 600mA to the existing z axis stepper motor, If I add a second stepper motor in series will I need to increase the milliamps?

If you have a free stepper driver you don't need to run anything in series.

        See here: https://duet3d.dozuki.com/Wiki/Bed_levelling_using_multiple_independent_Z_motors

If your Ender3 is largely stock I would say don't bother adding a second Z motor just because. The cantilever design is fine for the size and weight of the stock ender and adding a second z motor adds complication where it really doesn't need to. If you've got a heavy direct drive and you notice significant sagging and tilt that can't be corrected by adjusting tension on the wheel then it might be worth it.
